It was a crushing humiliation for French President Jacques Chirac, who had pulled out all stops in an attempt to have the constitution approved.
"The French people have given a huge smack in the face to an entire system that has the nerve to tell us what to think," said nationalist French politician Philippe de Villiers, an opponent of the charter. "The constitution is no more."
An estimated 55 per cent of a large turnout voted no in the referendum.
The rejection seems certain to be followed by another rejection in Holland on Wednesday and a no vote in Denmark in September.
The document needs to be approved by all 25 member states to survive.
"Realistically, this document seems to have been scuppered," said one diplomat in Brussels.
"How can it continue to exist when it has been rejected by the French, the nation at the heart of the EU since its inception?"
